Description
The LISBP (Laboratoire d’Ingénierie des Systèmes Biologiques et des Procédés) is a mixed research unit: INSA, INRA and CNRS with multidisciplinary activities on education, research and technology transfer in biotechnology and processes. It is a leading biotechnology laboratory in France, certified ISO 90001, which couples top quality teams in microbiology and protein engineering/biocatalysis with process engineering teams to enable a ‘gene to process’ approach.
The LISBP has longstanding and world renown in fermentation technology, biocatalysis and metabolic engineering (bacteria, yeasts and more recently microalgae) with a high level of transfer to industry, and an emerging reputation for systems biology, molecular evolution of enzymes, metagenomics screening, fermentation, separation and purification processes. More recently this overall expertise has evolved towards a more innovative approach involving a synthetic biology approach and a shift towards redesigning metabolic potential for ambitious biotechnology applications.
The LISBP has developed houses cutting edge platforms either post-genomic platforms with sequencing and transcriptomics facilities (Get-Biopuces), a metabolomics/ fluxomics RMN/MS platform (MetaToul), an engineering and screening for original enzyme platform (ICEO) or a fermentation platform facilitating the validation of industrial potential (FAMEtech).

GeT-Biopuces: is an integrated genomics core facility specialized in genomics and transcriptomics via microarrays and new generation sequencing (NGS). The platform is equipped with two array platforms: Affymetrix (GeneChip fluidic station 450, hybridization oven 640 and scanner 3000 7G with autoloader) and Agilent technology. In addition, a Qarray Mini spotter allowing to spot some oligonucleotides on glass slides to create your own microarray is available. The platform is equipped with an Ion S5 sequencer (Ion Torrent technology-ThermoFisher Scientific) and an Ion OneTouch 2 System performing template amplification and enrichment as part of the manual workflow for the Ion S5 system. The platform has a nanodrop 2000 to quantify nucleic acids and proteins as well as a bioanalyzer 2100 (Agilent) providing sizing, quantitation, and purity assessments for DNA and RNA.
MetaToul: State-of-the-art equipment in NMR (800 MHz & 500 MHz spectrometers) and mass spectrometry (LTQ-Orbitrap, Q Exactive +, QTOF Xevo, Q-TOF MaXis, Q-TRAP 4000) / Robotic plateform for High Throughput fluxomics (1 robot Evo200 for culture and extraction and 1 robot Evo200 for sample preparation
ICEO: Colony pickers, liquid handling stations specially developed for miniaturized and automated enzymatic assays, and continuously improved facilities for protein purification and characterization (HPLC, GC-MS, LC/MS equipments).
FAME tech: Highly instrumented bioreactors (1L to 20L) to carry out cultures of microorganisms in large operating conditions: batch, fedbatch or chemostat / Specific pilot configurations are developed to characterize either environment and/or biological behaviors and enhance basic knowledge / Micro-reactor (1ml) with the in situ measure control of pH, temperature and pO2 and in situ measure of optical density / Bioreactor for in situ mutagenesis / HDMB (High Dry Matter Bioreactor) an integrated bioreactor coupled a Twin-Screw Extruder for optimized implementation of microbial conversion at high lignocellulosic dry matter content. It physically combines the stages of physicochemical (reactive extrusion for lignocellulosic biomass pretreatment) following by enzymatic and microbial transformation of the released sugars under realistic industrial conditions in term of concentration and conversion / OSCAR: bi stage bioreactor with membrane recycling. This double reactor is intended to allow microbial cultures to be conducted in a controlled environment by disconnecting the residence time management for the liquid and biomass phases.

GeT-Biopuces: Genomic and Transcriptomic core facility (http://get.genotoul.fr)
Regroups both the expertise and the technology necessary for the analysis of gene expression at the level of entire genomes of different biological systems, notably that of micro-organisms using microarrays (Affymetrix-Agilent technologies) or NGS. Development of innovative technology for future generation of chip-based analysis. Support a wide variety of genomics projects on eukaryotic and prokaryotic genomes, even the most unusual ones (de novo sequencing or resequencing projects). Data analysis: quality control, normalization (R software and Bioconductor package) leading to differentially expressed genes analysis or functional enhancement. Analysis of NGS data. We provide theoretical and technical training.
METATOUL: Metabolomics-fluxomics (http://www.metatoul.fr)
METATOUL provides expertise and state-of-the-art methodologies in metabolomics and fluxomics for the functional analysis of metabolic networks with applications in health, biotechnology and microbiology. The platform focuses on quantitative approaches aiming at the quantification of metabolite concentrations (quantitative metabolomics) and of metabolic fluxes through 13C-labelling strategies. METATOUL develops innovative methods and tools aiming at the integration of all metabolic knowledge (metabolic systems biology).
ICEO: Engineering and Screening for Original Enzymes (http://cribligand.ipbs.fr/research.html#iceo)
ICEO technical and scientific skills are gathered to propose to the scientific community, both academic and from industries, a large set of services, in the field of enzyme discovery and engineering. The platform provides expertise in screening of micro-organisms (bacteria, yeasts, fungi,…), exploration of natural (genomic or metagneomic libraires) or engineered (mutagenesis, directed evolution) diversity. ICEO develops new and original screening methods, specifically adapted to the targeted function (specificity, stability, catalytic efficiency, solubility, expression level, affinity,…)
The expertise of ICEO covers 2 main domains:
Creation of diversity:
- construction of genomic or metagenomics libraries,

- in silico engineering,
- rational / semi-rational engineering.
Enzymatic activity screening:
- setting up selection tests,
- miniaturization of tests for HTS,

The objective of this platform is to emerge innovative technologies for the intensive production of molecules for energy or chemical compounds from renewable carbon and industrial by-products to formulate molecule/microorganism/process solutions adapted to customary constraints. Set up and development of methodologies and tools to follow and characterize (online, off line) microbial cultures associated to nutritional strategy in large operating conditions: batch, fedbatch or chemostat, synthetic or industrial media, mono or co-substrates, pure or co-cultures in axenic conditions…
